After upgrading from 1.5 to 1.6.6, the Global Settings link in the admin menu gives me the following error:
string(54) "Smarty error: unable to read resource: "siteprefs.tpl""
The file existed, but was blank. I replaced it with a siteprefs.tpl file from another 1.6.6 install. Still doesn't work. Any ideas?

Have you tried running a checksums on your file to ensure the integrity of all your files.
Maybe try downloading the package again and reuploading the siteprefs.tpl file again.

The checksum reported over 100 files missing. I reverted back to my 1.5 install re-upgraded to 1.6, then 1.6.6. It now works fine. One thing I did differently - I uninstalled the news module before upgrading. News has been causing me some problems, so I thought 'why not?' Don't know if that made the difference, but it worked.

Shouldn't have affected it, probably just a bad FTP session or something like it.
