After a fresh install of Version 1.10 beta 3 I keep getting an unhandled notification.

CMSMailer Your mail settings have not been configured. This could interfere with the ability of your website to send email messages. You should go to Extensions >> CMSMailer and configure the mail settings with the information provided by your host.

I have configured CMSMailer and the unhandled notification is still there.

This behavior is afaik a symptom of the extended use of pseudocron routine that checks the system for new versions etc.
So if the site is active this message should disappear automatic after about one hour (if settings for pseudocron is not changed from default settings).
I agree its confusing that the message is not removed at once....

Just check my (test) site again and the unhandled notification is gone, as you wrote it would be. Is, indeed a little confusing.
